New Confidence: Optimizing Benchmark Dose Modeling
to Improve Risk Assessment

چکیده

Benchmark dose (BMD) modeling has been touted as a more accu rate method for conducting chemical risk assessments. In this issue of EHP, a team of researchers applied BMD modeling to hundreds of chemicals whose health effects are already known in an effort to optimize the approach. To calculate the highest safe oral dose of a noncarcinogenic chem ical, regulators must first estimate the pointofdeparture (POD) dose—that is, the dose below which the chemical does not increase disease risk. This POD is then divided by an uncertainty factor to build in extra safety, resulting in a reference dose. In selecting a POD regulators may use the lower confidence limit of the BMD (a value abbreviated BMDL), or they may use the noobservedadverseeffect level (NOAEL) obtained from animal experiments.

Background and aims: Exposure to the normal hexane can induce dysfunction of the nervous system and color vision impairment. Occupational risk assessment is established according to no observed adverse effect levels (NOAEL). Recently, because of the NOAEL’s limitations, the benchmark responses were applied for risk assessment. The study’s aim is an analysis of benchmark response in occupational...

Simultaneous lower confidence bounds on benchmark dose profiles for joint-action continuous data

Benchmark analysis is a widely used tool in quantitative risk analysis. A core operation therein is estimation of minimum exposure levels, called Benchmark Doses (BMDs), that induce a pre-specified, adverse, Benchmark Response (BMR) in the target population or system. This approach is well understood when a single hazardous stimulus is studied. Deutsch and Piegorsch (2013) expand the estimation...

Estimation of the benchmark dose by structural equation models.

While epidemiological data typically contain a multivariate response and often also multiple exposure parameters, current methods for safe dose calculations, including the widely used benchmark approach, rely on standard regression techniques. In practice, dose-response modeling and calculation of the exposure limit are often based on the seemingly most sensitive outcome. However, this procedur...

Epidemiology-based risk assessment using the benchmark dose/margin of exposure approach: the example of ethanol and liver cirrhosis.

BACKGROUND A novel approach to derive a threshold dose with respect to alcohol-related harm, the benchmark dose (BMD) methodology, is introduced to provide a basis for evidence-based drinking guidelines. This study is the first to calculate a BMD for alcohol exposure using epidemiological cohort data. With this BMD we will be able to calculate the margin of exposure (MOE) for alcohol consumptio...

A Signal-to-Noise Crossover Dose as the Point of Departure for Health Risk Assessment

BACKGROUND The U.S. National Toxicology Program (NTP) cancer bioassay database provides an opportunity to compare both existing and new approaches to determining points of departure (PoDs) for establishing reference doses (RfDs). OBJECTIVES The aims of this study were a) to investigate the risk associated with the traditional PoD used in human health risk assessment [the no observed adverse e...
